The 6xHis tag is a commonly used protein purification tag in molecular biology and biochemistry. It consists of a short amino acid sequence containing six consecutive histidine residues (His). This tag is often genetically fused to the target protein of interest, allowing for purification of the fusion protein using affinity chromatography.

The purification process typically involves the use of a metal ion affinity resin, such as nickel-nitrilotriacetic acid (Ni-NTA) agarose, which has a high affinity for the His-tagged protein. The His-tag binds specifically to metal ions immobilized on the resin, allowing for selective purification of the fusion protein from a complex mixture of proteins.

After purification, the His-tag can sometimes be removed from the target protein through enzymatic cleavage or chemical methods, leaving behind the native protein. The 6xHis tag is widely used due to its small size, high affinity for purification resins, and minimal interference with the structure and function of the target protein.
